Perform research to study transcription and/or DNA properties that affect transcription and its regulation using techniques the best suited single molecule techniques available in the lab, which span from TPM, atomic force microscopy (AFM), magnetic tweezers and the C-trap microscope. Acquisition of data and data analysis. Draft manuscript describing results. Other duties as assigned
JOB DUTIES: 40% - Essential - Use single molecule techniques to investigate DNA transactions, including (but not necessarily limited to) transcription, compaction by regulatory proteins, conformational changes as a function of the environment. These investigations involve preparation of (i) microchambers to look at samples under the microscope, (ii) DNA-protein samples, (iii) manipulation of DNA-protein complexes applying torsional and/or tensile forces, (iv) data acquisition and analysis, (v) preparation of figures summarizing the experimental data collected, (vi) limited coding, (vii) writing drafts of paper and (viii) possibly presenting results at meetings.


20% - Essential - Prepare samples needed for single molecule microscopy. These preparations involve the assembly of microscope chambers from slides and coverslips, preparation of buffers and basic molecular biology such as pcr, restriction and ligation reactions.


20% - Essential - Acquisition of data and data analysis. This may involve recording of AFM images, single particle tracking in TPM, magnetic and optical tweezers and/or quantification of fluorescence intensity in the C-Trap. Data analysis will involve plotting of data, applying physics and mathematical modeling, with possible tuning of existing codes or writing of new algorythms and interpretation of the results.


20% - Essential - Draft manuscript describing results will involve preparing figures and writing text that introduces the significance of the research, describes the materials and methods, describes the results and discusses their meaning.
MINIMUM REQUIREMENTS: Education - Doctoral / Professional Degree
Experience: Research at the doctoral and postdoctoral level, single molecule expertise

COMPENSATION INFORMATION Expected Salary Range: $42,800-$60,000

Salary is dependent upon several factors including, but not limited to, a candidate's previous experience, knowledge, skills and performance in accordance with Clemson's compensation guidelines.

ESSENTIAL PERSONNEL LEVEL Normal Operations


Required to follow emergency facility closure directives, and not normally expected work on-site during emergency situations.
